AFAIK, only global filtering does that. However, it cannot alter messages. If the server you forward to can be found via MX you need no other setup.

may be can we run two intances of courier or can we differentiate between local delivery and smtp and forward the mails to another server if it is a local delivery.

If you want to deliver locally without defining local mailboxes, you can install a .courier-default in aliasdir. If you do that, then you _must_ set your maildropfilter file in order to check that the remote mailbox exists, e.g. http://www.courier-mta.org/localmailfilter.html
