I am using round-cube web mail for my setup(exim, courier, mysql). I manually
created maildirsize with the command (maildirmake -q 100000000S ./Maildir).
After that  quota shows but not updated automatically i need to manually hit the
same command and quota get updated, Is there any other method to update maildir
quota in courier.

Whatever process you use to deliver mail to the maildir is responsible for
updating the quota, in your case it's exim.
