I tried releasing the lock this way, but I can not find any "home directories" which are +t. vchkpw runs (I believe) as the "vpopmail" user, and it's home directory is /usr/lib/vpopmail, which is not +t. The server runs as root, which is not +t. What am I missing here?

This would be the home directory of the virtual user, as defined by the vpopmail driver. This will be either the maildir directory itself, or its parent directory.
