I have maildrop setup with a simple config to check mail using spamassassin. What I haven't been able to figure out is how I can send back an error to the sender if spamassassin's says the mail is spam, instead of delivering the mail. Is there a way to generate a 550: this is spam or something from maildrop?

Depends on how maildrop is being called of course -- if its being called before the SMTP connection is gone from your mail server then yes, it could. If the message is "delivered" to maildrop after the SMTP connection is closed then the sender already has their 200 response.

Remember not to bounce messages manually (replying to header senders, etc.) if they're suspected of being spam since those headers are likely forged.
