It looks like you're just relaying mail for the "dsmedialabs.com" domain, right?

Deliveries are failing with "DNS lookup failed." You need to resolve the DNS issues so that delivery works. Once courier is able to deliver mail, it'll stop refusing new messages.

Oddly i have 127.0.0.1 mapped to dsmedialabs.com in the hosts file is this not enough? do i need to have it in DNS?

Yes you do. Courier always uses DNS.
