One more thing: I compiled everything (maildrop, courier-authlib, courier-imap, postfix, openssl, etc) with the following FLAGS:
CXXFLAGS=-march=nocona -O2 -pipe -fomit-frame-pointer -mmmx -msse -msse2 -msse3 CFLAGS=-march=nocona -O2 -pipe -fomit-frame-pointer -mmmx -msse -msse2 -msse3

First of all: Should I post the questions in the courier-imap list or courier-users list ? :-)
Just to avoid double posts.

I use postfix. in the master.cf:
maildrop unix - n n - - pipe flags=DRhu user=maildrop argv=/usr/local/maildrop/bin/maildrop -d ${user} -w 90
Run maildrop -v to verify that quote extensions have been compiled in.
~# /usr/local/maildrop/bin/maildrop -v maildrop 2.0.3 Copyright 1998-2005 Double Precision, Inc. GDBM extensions enabled. Courier Authentication Library extension enabled. Enabled Berkeley DB instead of GDBM extensions. Maildir quota extension enabled.
What about your filesystem, and mail delivery rate?
Distribution: Ubuntu 6.10 server
Kernel version: 2.6.20.3 (vanilla compiled by hand)
Filesystem of Maildirs: XFS # grep "/home" /etc/fstab /dev/sda3 /home xfs noatime,nodev,noexec,nosuid 0 2
maildrop permissions: # ls -l /usr/local/maildrop/bin/maildrop -rwsr-sr-x 1 root mail 195217 2007-03-08 12:23 /usr/local/maildrop/bin/maildrop
About 400 concurrents users acessing by imap. There is a perdition proxy in other server before courier-imap in this server.
Under normal conditions, every once in a while -- but vary rarely -- it is possible for a quote update to get missed. That's normal, but shouldn't occur too frequently.
Can I force to calc the used quota for a single user, without send mail to him, by example ?
I use horde imp imap client for deletion of messages and Mozilla Thunderbird.
That does not answer my question. I did not ask which IMAP clients you are using, but how they implement message deletion in IMAP.
I don't know how to answer this question :-(
Neither can I, only you can answer this question. You'll have to set up a test mailbox with a test message, delete the message, and see what's in the maildir.
