/home/vpopmail/domains/domain1.com/user/Maildir

Yes, the INSTALL notes indicate that you may have to manually edit DEFAULT_DEF.

However, maildrop does not support vpopmail's mail storage hierarchy. There is no appropriate DEFAULT_DEF setting for vpopmail.
