Dear Sam ,
I found something special that's may be a hints could find out what
cause the maildrop can't be run .mailfilter on my machine.
please take a look the test shown as below.
Test 1:
cat /etc/issue | sudo -u vmail maildrop -V -d data...@test.com
maildrop: Unable to open filter file, errno=2.
Test 2 :
cat /etc/issue | sudo -u vmail maildrop -V 6 -d data...@test.com
< result is all right >
for these two test. .mailfilter also locate in
/home/vmail/domains/.mailfilter
By the way . What's the different between -V 6 & -V under CMD enviorment ?

Well, you have apparently proven that if you invoke maildrop correctly, the
right thing happens. Therefore, the logical conclusion is that maidrop is
not being invoked properly, by Postfix. It's probably some configuration
issue, so it sounds like running your Postfix config by the postfix mailing
list is the best way to find out what's not set correctly.
